Confocal Laser Scanning Microscopy (CLSM) is an optical imaging technique that is comparable in quality to scanning electron microscopy (SEM) at the same magnifications, with the addition of a spatial pinhole to focus a laser beam on a specific position on the sample. As only one position in the sample is illuminated at any one time, more time is required to scan across the entire sample, thus creating better contrast. The confocal microscope is also equipped with a TFT touchscreen monitor and motorized light control which allows z-stack scanning with a 10nm increment, thereafter going through processing to create a 3D image. The Time Series module allows users to compile images of specimen captured over time. Four diode laser of laser class 3b are available: 405nm, 488nm, 561nm and 640nm. Offline workstation installed with “ZEN 3.0 (blue edition)” available for user to post process, edit and analyse images.
The laser source of this system is completed enclosed by an opaque barrier. Thus, the user of this system is exempted from obtaining the N3 license.
What kind of research is applicable / suitable for this equipment?
- Users who wish to obtain high quality images or structures of specimens in their natural 3D state
- Users who wish to observe and reconstruct in detail the habitus and dissected appendages, whole specimens and any dissected parts
- For long-term digital records purposes. Photos taken by CLSM reduce the need of acquiring type specimens from museums for comparisons, which often damage type material in the midst of transfers and lab activity.
